The way I normally do it (with Professional Acumen which reduces the first-time use cost of an assassin ability each round):

  1. Either charge into the enemy (if you have 6 AP or the boots that reduce the charge cost by 1) or simply walk into the melee distance from the direction of the opening.
  2. Buff time: Reckless Strike, Sworn Enemy, Sure-Fire Plan (if you chose the Crime Lord origin).
  3. Main hand attack into the opening.
  4. Create a second opening with your assassin skill, reuse Sure-Fire Plan (if you have it).
  5. Off-hand attack into the opening.
  6. Death Whisper (free, 0-AP attack for 25% of the damage).

When an officer gives you an extra turn with 2 AP, you can charge into a different enemy (if you have the charge boots), main hand attack + Death Whisper. Or stay on top of your current target and simply do main hand + Death Whisper.

If the enemy is at half health or lower and you have a decent hit chance, replace one of the attacks with Killing Edge for the ultimate damage.

Feats that help to increase the damage: Desolation and anything that adds extra Direct Damage on top of your normal attacks. There is a feat that gives +1 Direct Damage for every 10 damage you do, and multiple feats that increase damage to targets at full health based on lethality – take them all.

I use power weapons and Drukhari swords on my assassin to ignore as much armour as possible.

Stack Weapon Skill, Agility and Perception to increase your dodge, dodge reduction and crit chance as much as possible. Strength can be ignored until later, when you have spare points for it.
